Item Identification of Pathogenic Bacteria Isolated from Fingernails of Children in Shendi Locality(جامعة الشيخ عبدالله البدري, 2022-03-09) Lana Jamal Abubaker; Mosab Nouraldein Mohammed Hamad; Tarig Mohamed Ahmed Mohamed; Khalid S Hammad1; Ghanem M Mahjaf; Leila M AbdelgadeirAbstract Background: Recently a lot of studies focused on the global occurrence of resistant bacteria carriages. Fingernails are a perfect area for harboring bacteria. Children are the most age categories who harbor pathogenic bacteria in their fingernails. Objective: This was a prospective cross-sectional descriptive study aimed to identify the pathogenic bacteria isolated from the fin gernails of children in the Shendi locality; it also studied their antimicrobial susceptibility patterns. Methodology: The study included (70) participants, 37 (53%) males, and 33 (47%) females, within the age group (1- 10) years. Nails were collected and firstly cultured on nutrient broth then on blood agar and MacConkey incubated aerobically at 37c° for 24 hours. Isolated organisms were identified according to their colonial morphology, indirect gram staining reaction, biochemical tests, and antimicrobial susceptibility tests were done by the disc diffusion method. Result: The study revealed that 61 (87%) of children showed bacterial growth from fingernails. The isolated bacteria were Staphy lococcus aureus18 (29.1%), Bacillus spp 11 (17.7%), Escherichia coli 2 (3.2%), Klebsiella spp12 (19.3%), Enterobacter spp8 (13%), Citrobacter spp6 (9.6%), Proteus vulgaris4(6.5%) and Salmonella spp1 (1.6%). Ciprofloxacin 58 (93.5%) and Gentamicin40 (64.5 %) were the most effective antibiotics against the isolated bacteria in contrast they were mostly resistant to Nitrofurantoin 53 (85.5%), Amoxiclav 45(72.6%) and Doxycycline 39 (63%). Conclusion: Long nails and playing behaviors of children were the main factors for harboring bacteria in fingernails. Staphylococcus aureus was the most isolated bacteria while Ciprofloxacin was the most effective antimicrobial agent used.Item In vitro Assessment of Antimicrobial Activity of Honey Bees and Nigella sativa Against Selected Clinical Isolates from Shendi City(جامعة الشيخ عبدالله البدري, 2022-03-16) Ghanem M Mahjaf; Leila M Abdelgadeir; Aisha Y Ibrahim; Nedal A Mohammed; Sulafa A Mohammed; Salih O Mohammed; Mosab Nouraldein Mohammed Hamad4; Naser E BilalAbstract Background: An alarming rise in bacterial strains resistant to existing antimicrobial agents requires a renewed effort to seek agents effective against pathogenic bacteria resistant to antimicrobials. The new antimicrobial agents are will overcome this problem. Objective: This study aimed to study the in vitro antimicrobial activity of different concentrations of Honey, petroleum ether extracts of medicinal plant Nigella sativa (seeds), and the mixture using the cup-plate agar diffusion method. Materials and Methods: This cross-sectional observational study was carried out among One hundred samples were collected from urine, wound, ear, and eye swab, 95/100 (95%) showed bacterial growth, from which six types of pathogenic bacteria at Shendi city hospitals during March to June 2018. Results: Of the total 95 clinical and standard specimens were confirmed as Staphylococcus aureus 20 (21%), Escherichia coli 20 (21%), Pseudomonas aeruginosa 15 (16%), and Klebsiella pneumoneae 30 (31%). In addition to Candida albicans 10 (11%). These results showed the activity of antimicrobial Petroleum ether extract of the Nigella sativa, and honey pronounced dose-dependent on standard strains and clinical isolates, while methanolic extract of Nigella sativa explained no activity. Conclusion: The activity of Nigella sativa, honey, and the mixture exhibited high antimicrobial activity against all types of tested or ganisms both clinical and standard organisms. Therefore Nigella sativa, honey, and the mixture can be regarded as a broad-spectrum antimicrobial agent.
